(1)细胞对凝血系统的调控机制,及其在重大疾病中的作用

探索血液循环中的细胞(血小板、红细胞、白细胞、胞外囊泡、癌细胞等)对凝血级联系统以及纤溶系统的新调控机制,以及此类调控机制在重大疾病如心血管疾病和癌症中的作用。

(2)新型凝血功能检测法的开发和应用

凝血酶生成检测法 (Thrombin generation assay, TGA)是目前最广泛应用的新型凝血功能检测法,它能提供比传统凝血检测法(PT、APTT)更高的灵敏度。本人已成功开发新型全血凝血酶生成检测法(全血TGA),可对人和鼠全血样本的凝血潜能进行高灵敏度的直接量化检测。       全血TGA优点包括:(1) 直接检测全血样本,省去了对样本进行预处理的时间,有潜力开发成为床旁检测法(POCT)(2) 全血TGA相比血浆TGA的灵敏度更高:能更灵敏的检测出小鼠凝血因子IX, XI, PKa C1-INH的敲除对凝血系统的影响。      全血TGA潜在应用领域: (1)作为研究红细胞等血细胞对凝血过程的调控机制的新工具, 为临床血栓/出血疾病提供机制解释;(2)高灵敏度地筛选新型抗栓/止血药物(有效性和特异性); (3)优化/指导临床抗栓/止血药物的剂量; (4)作为指示临床血栓/出血风险的POCT。

(1)细胞对凝血系统的调控机制,及其在重大疾病中的作用
